Abstract (EN): Our aim was to investigate the effect of high glucose exposure on the absorption of folate (FA) by Caco-2 cells. We verified that apical (AP) high glucose (both different concentrations (10-45 mM) and different exposure times (10 min-24 h)) did not affect the AP uptake of H-3-FA. Also, AP high glucose (30 mM) did not affect the intracellular steady-state levels of H-3-FA, and simultaneous AP and basolateral (BL) high glucose (30 mM) did not change the AP-to-BL apparent permeability (P-app) to H-3-FA. The effect of several drugs (5-methyltetrahydrofolate, methotreXate, SITS, DIDS, indomethacin, p-aminohippuric acid and furnitremorgin C) on H-3-FA uptake or P-app was not changed or only hardly changed by high glucose. We conclude that absorption of H-3-FA is not modulated by either AP or BL high glucose exposure.
