Os receptores do glutamato têm sido implicados em diversos fenómenos fisiológicos e patofisiológicos. Entre eles, os receptores metabotrópicos do glutamato (mGluR), têm recebido particular atenção durante os últimos anos devido ao aparecimento de dados sugerindo o seu envolvimento no processamento da informação nociceptiva. O presente trabalho teve como objectivo o estudo da expressão do ARN mensageiro (ARNm) para diferentes subtipos de mGluR no sistema somato-sensitivo do Rato, e eventuais alterações daquela expressão induzidas pelo aumento crónico de afluxo nociceptivo. Previamente, fez-se uma determinação da actividade metabólica neuronial, por injecção endovenosa de 2-deoxiglucose radioactiva e detecção autorradiográfica, para determinação de algumas das áreas do Sistema Nervoso Central (SNC) que se encontram activadas, e do respectivo padrão temporal de activação, por aumento do afluxo nóxico decorrente da indução de uma monoartrite. A técnica de hibridização in situ foi depois utilizada para se avaliar a expressão do ARNm de diferentes mGluR nos animais monoartríticos, em algumas das áreas onde ocorreram alterações da actividade metabólica neuronial. Particular atenção foi dada ao complexo ventrobasal talâmico (VB) e núcleo posterior do tálamo (Po), responsáveis pela transmissão da informação dolorosa ao córtex somato-sensitivo, e ao núcleo reticular do tálamo (Rt), importante na mediação dos impulsos do circuito tálamo-cortico-talâmico. Neste último núcleo, procedeu-se ainda à injecção estereotáxica em ratos monoartríticos, de um antagonista específico do grupo II dos mGluR, para se avaliar o papel destes no processamento do afluxo nociceptivo nesta região.
Abstract (EN):
Glutamate receptors have been implicated in several physiological and patophysiological mechanisms. Among them, the metabotropic glutamate receptors (mGluR), received a particular interest more recently since several data appeared suggesting they participate in the processing of noxious input. The present work was devised to study the expression of mRNA for different subtypes of mGluR in the somatosensory system of the Rat, and eventual changes of that expression during the development of chronic inflammatory pain. Previously, neuronal metabolic activity was measured, by endovenous injection of radioactive 2-deoxyglucose and autoradiographic detection, to determine some of the Central Nervous System (CNS) areas which are activated by the increased noxious input arising from monoarthritis induced by Complete Freund’s Adjuvant. Additionally, the temporal pattern of that activation was also analysed by using monoarthritic rats at different time points of evolution of the disease. The expression of different mGluR mRNA in monoarthritic rats was then studied in some of the areas showing alterations of the neuronal metabolic activity, by using in situ hybridization with oligonucleotide probes. Particular attention was given to the ventrobasal thalamic complex (VB) and posterior nucleus of the thalamus (Po), relay nuclei which transmit the noxious information to the somatosensory cerebral cortex, and to the reticular nucleus of the thalamus (Rt), which has an important modulatory role in the thalamo-cortico-thalamic loop. In the Rt of monoarthritic rats, we have also performed stereotaxic injections of a specific antagonist of group II mGluR to evaluate the role of these receptors in the processing of nociceptive input in this thalamic region.
