Abstract (EN): Total serum alkaline phosphatase (AP) activity reflects individual isoenzymic activities. Characterization of AP isoenzymic pattern provide useful clinical informetion. Various experimental conditions for identification and quantitation of rat serum AP electrophoretic fractions were studied. Electrophoresis was performed on cellulose acetate using, for each sample, two distint buffers, sodium dietylbarbiturate (veronal)HCl (Ver) and 2-amino-2-methylpropan-l-olHC1 (AMP) and also a mixture of both (AMP+Ver), pH 9.4 andor 10.4, and two substrates (- and 8-naphtyl phosphates). Animals (n=8) were fed ad libitum. Two serum electrophoretic fractions bands were separated and their percentage values and calculated activities were analysed. Independently of substrate: 1)the adition of AMP to Vet significantly modifies the percentage values of both fractions but the addition of Ver to AMP does not induce any significant change; 2)significantly different percentage values were obtained with different pH. Calculated activities of each fraction were significantly modified with: 1)the addition of AMP, except with -naphtyl phosphate for the first fraction; 2)the addition of Ver for -naphtyl phosphate; 3)pH change, except for second fraction with -naphtyl phosphate. In conclusion: 1)the presence of AMP or pH 10.4 significantly increase first fraction percentage; 2)AP fractions assay depends on technical conditions.
