Abstract (EN): Diploid germ cells (DGC) were isolated and cocultured for 2-weeks in Vero-cell conditioned-medium (CM). FSH and Testosterone (T) elicited 7% of meiosis (new round spermatids, Sa1) and 17% of late spermatid (Sd) differentiation. Developmental arrest occurred at early (Sa1Sa2) and late (Sb-Sd) spermiogenesis. Transmission electron microscopy (TEM), fluorescent in situ hybridisation (FISH) and mRNA expression (RT-PCR) confirmed cell-stage diagnosis and normal meiotic chromosomal synapsis and segregation. DNA replication (BrdU) of DGC was kept for only I week. Cultures were limited by apoptosis, as confirmed by caspase-3 activity, TEM and RT-PCR.
