Abstract (EN): DYS391 and DYS393 primers, described in the Genome Data Base, were used to amplify DNA samples from both male and female origin. After the amplification of DNA from male blood samples it was possible to observe additional amplification products to the normal alleles for both systems when an annealing temperature of 55 degrees C was used. In addition, in the same size range a clear polymorphism was detected in females for both systems. For DYS391, a first screening in males on these minor amplified products allowed the identification of 5 different alleles. After the amplification of male DNA samples only single band patterns were observed while using the same conditions to amplify female DNA it was possible to observe the same polymorphism but with the presence of heterozygous patterns, as expected for a diploid genetic system. In order to improve the applicability of this system in forensic genetics, an alternative primer was designed to amplify the Y-chromosome specific STR.
