Abstract (EN): We explore a simple approach to carry out fluorescence in situ hybridization (FISH) on-chip: combining on-chip magnetic capture of Escherichia coli based on immiscible-filtration-assisted-by-surface-tension (IFAST), followed by detection of the isolated E. coli with FISH in the same device. Preliminary experiments show successful pathogen capture followed by a partial on-chip FISH assay using 107-109 cells mL-1 fixed and permeabilized in three conditions: (1) 4% vol/vol paraformaldehyde and 50% EtOH, (2) 50% EtOH, and (3) 100% EtOH. © 2019 CBMS-0001.
